Yes bad throttle shaft seals/bushing etc will cause on to run rough and if bad enough keep it from running at all but it would have to be real bad. That is why all the kits I have ever seen have seals for the shafts in them. I have yet to find many carbs that can not be rebuilt unless there are broken parts that is. As for only running with your hand over the end of the carb means you are not getting enough fuel to the carb or to the engine. I would start by pulling the drain plug out of the bottom of the carb and let the fuel flow for at least 5 minutes. Catch that liquid gold so you can look at it for water and dirt etc. If you do not have a good flow work your way back till you find where the flow stops and fix it
