1995 f250 tranny

I had two F250 diesels. Both 4WD, extended cab, and 4.10 axle ratio. Pretty much exactly the same except one had the C6 and the other has the E40D. C6 got 13-14 MPG on empty highway trips. E40D gets 16-17 MPG I assume due to lock-up converter and overdrive. When worked hard hauling heavy trailers - they got the same MPGs.
 
You might lose a little with an auto tranny, but I've never had a truck with an auto yet. My current truck is a 95 F250, 5.8 w/the manual overdrive, 2wd. I can get 14 mpg with mixed driving. The best I've gotten was 14.5. Doesn't really get much better straight highway mileage, but it seldom sees straight highway without some sort of a trailer behind it. Road trips are what the family hauler are for...

Only time I like a auto tranny is stop and go traffic.
I have a 4 speed plus OD in my 87 F150 with 5.0L and 3.55 gears front and rear.
Get 14.5 MPG with mostly highway driving.
 
I ordered out a brand new '87 F-150 4wd, 300-6, creeper low NP-435 4-speed, 3.55 gears, and 14.5 mpg is about all the better mpg it would get also. Buzzing that 6 cylinder to 2600 rpm to run 65-70 hurt right in the wallet buying gas. Truck was a reg cab short box, 117 inch wheelbase, about 34 gallon gas capacity with dual tanks, had to drive easy to get over 450 miles on both tanks. No tilt, cruise, or AC.

Getting my '96 F-250 diesel was best thing I did. 18-19 mpg at 70-75 mph. 800 miles between fuel stops. Went three months one summer, 10,000 miles and never dropped below 20 mpg.
 
Neither c6 nor 4speed have od. That said a c6 has torque converter slip and a four speed didn't. the four speed will get better place.
 
You will get the best mileage with an overdrive whether it is in a manual or automatic. Today's automatics have locking torque converters, so there is no significant slippage during highway driving. However, bear in mind that modern transmissions are electronically controlled and need to have the correct processor to powertrain match. Otherwise, there will be nothing but troubles - like MIL constantly lit, trans might not shift or hold properly, or a whole host of other problems.

The 4 speed automatic in those trucks, the E4OD, had overdrive, the C6 was a 3 speed.

The 4-speed manual did not have OD, needed the ZF5 5-speed for OD.
