450 ignition timming

Do I use the 5th mark on the crank or the first mark? Also once that is determend on the pointer does that mean that the #1 cylinder is at top and the rotor button arm is pointing to #1 cylinder. Before I can start fine tuning ?

Static timing (non running) should use the last mark in the direction of rotation. TDC.
It is TDC on compression 50% of the time. 4stroke has the end of exhaust and beginning of intake on TDC as well. Put your thumb in the #1 plug hole and crank/move the engine till it starts to blow air out the hole (compression stroke) If timed on the exhaust stroke it will be 180 off. JimN
