If the rims are on the cast now, I would put them on together.

If they are apart, I think it would be easier to put the cast on first then put the rims on while they are vertical.

The cast centers are heavy and if mounted to the tire/wheel make it very difficult to move around. It is possible to put them on together but I find it easier to mount them separately.
 
Engine hoist, cast, then rims. I tipped mine over with a much lighter C wheel with rim attached. That is when I started removing the rim.
 
I have only done mine as one unit and I can't imagine it being any easier than doing it that way.
Note - When moving tires tell your buddy to back the heck off and get another beer and DO NOT try to help. They are always easier to move by yourself.
